Comparison of specific [ 3 H]OXT binding to wild-type and mutant oxytocin receptors. COS-7 cells were transiently transfected with empty pcDNA3.1 vector and vectors encoding wild-type and mutant OXTRs, and were exposed to 50 nM [3H]OXT in the absence and presence of 20 μM unlabeled oxytocin. Specific binding was determined as described in Methods. Statistical significance of differences in [3H]OXT specific binding between wild-type- and mutant-transfected cells was determined using one-way ANOVA followed by Dunnett's test. Data presented are mean in cpm/106 cells ± SEM of multiple experiments performed in duplicate. * p<0.05.
